This is the 21st century update on the acronym S.H.I.E.L.D. although not actually spoken until the end of movie (Iron Man).

The running gag is, "I'm Agent Coulson with the Strategic Homeland Intervention, Enforcement and Logistics Division, can we have a word with you" responding person, "Strategic Homeland Inter-whats... you guy have to come up with a shorter name" Agent Coulson " Yeah, we get that a lot."

At the end of the movie: Pepper Potts, "Can you thank the Strategic Homeland Intervention, Enforcement and Logistics Division for us?" Agent Coulson, "Just call us SHIELD."

SHIELD was developed during WW2 when spying on each other was what kept most people alive. The acronym originally stood for Supreme Headquarters, International Espionage, Law-Enforcement Division.

After the chemical attacks and subsequent invasion of Kuwait by Iraqi forces, it was changed in 1991 to Strategic Hazard Intervention, Espionage Logistics Directorate.

In 2008's Iron Man film, the acronym stood for Strategic Homeland Intervention, Enforcement and Logistics Division. Aside from actually putting the letters together various hints are given about SHIELD's presence in the film, such as the actual logo on one of the computers, along with the words spelled out and first letter capitalized.
